 You can use a pickup line if you make it blatantly obvious that it's a joke, and you're not taking it seriously.
A better plan, especially for the scenario you've listed, is to say Hi, ask her about her weekend, make a joke about it, then ask if she's doing anything on X day. If she says she has plans, offer up one more alternative. If she says she has plans again, without offering her own alternative, it's not going to happen, so shrug and just keep talking a few minutes before leaving.
If the conversation goes well, you can delay the asking her out part, but the whole exchange should take less than five minutes.
Most importantly, be prepared to get shot down. Go into this accepting that chance, and being at peace with it. Few things are a bigger turn-off to chicks than seeing a guy who cannot handle a "No." It's not the end of the world. Good luck.

There's really no such thing as a good pick-up line. There a player's lines, and corny lines.
My advice if you really like this girl is to just tell her that you really like her, and would like to ask her out. That gets to the point, so she knows you're serious, and it makes sure you don't end up sounding corny or something. Tell her how you feel, from your heart, not your head. If you fancy her, tell her that you fancy her and would like to get to know her more.
