 Well, it's usually pretty clear cut when a girl says she thinks of a guy as a big brother. it's usually that she's connected with you on another level rather than an intimate one. If she's already stated that's how she feels about you, I'm not quite sure if there's much else for you to do. You can still tell her how you feel, but just be prepared that it may not turn out how you would prefer.
